Investigating the molecular mechanisms of the effects of Capparis ovata extract in experimental animal model of multiple sclerosis.

Özet:

Multiple sclerosis (MS) is an autoimmune inflammatory disease of central nervous system (CNS) that contains demyelization leading patches of plaques formation in CNS. Caper parts were supplied by Asci Murat Capers, Ice Cream, Dessert and Pickle Manufacturing & Export Co., Ltd. Caper has been widely used by the traditional medicine for healing effects in certain pathological conditions. The aim of the present study is to determine the possible therapeutic effect of Capparis ovata extract an experimental model of MS. Experimental autoimmune encephalomiyelit (EAE) was induced in six to eight-week-old female C57 BL/6 mice by the procedure which had been described previously. Mice were randomly assigned to two groups: EAE control mice and caper extract-treated EAE mice. Caper extract-treated EAE mice, received 500 mg extract per kg body weight intragastrically for 21 days while EAE control subjects received only water. Animals were killed by decapitation and brains were removed. Total RNA was isolated using ?Qiagen RNeasy Lipid Tissue Mini Kit? in accordance with the manufacturer?s standard protocol. RT-PCR was performed using the First-Strand cDNA Synthesis System following the manufacturer?s protocol. We have observed the same levels of mRNA expressions for 2', 3'-cyclic nucleotide 3'-phosphodiesterase (CNP), myelin associated glycoprotein (MAG), myelin basic protein (MBP), matrix metalloproteinase-2 (MMP-2), NF-?B p50 and p65 sub unit, proteolipid protein (PLP), peripheral myelin protein (PMP) and tumor necrosis factor ? (TNF-?) studied in both Caper extract-treated mice, EAE control mice, caper extract fed- and control- mice. While CNP, MAG, MBP, PLP and PMP mRNA level are decreased to compare control, Caper group are returned control level. In addition, MMP-2, NF-?B p50 and p65 sub unit and TNF-? expression levels was increased to compare control. Caper treatment group was decreased. This result shown that caper extract may be therapeutic and protective effect for MS.
